Why I had to reverse genetically modify e.coli for my gene therapy

I ran into some strange issues while moving the open source gene therapy / Follistatin project from version 6 to version 8. I ended up having to cure my e.coli of their plasmids to solve it. This covers the plasmid curing process and some trouble shooting methods I used.
